【摘要】

Disruption of cell-matrix interactions is associated with a wide range of diseases such as cancer, fibrosis, and neurodegenerative diseases. Modified interactions have major effects upon key functional cellular features, as well as upon the matrix biophysical and biochemical properties. In this talk, I will focus on my recent work of improved characterization of these structure-function connections based on endogenous contrast of cells and tissues. With regards to the cellular features, we use two-photon excited fluorescence (TPEF) microscopy to acquire images from conenzymes of NADH and FAD which are actively involved in a number of metabolic pathways, and develop novel optical biomarkers to characterize cell features, including the optical redox ratio, phasor fluorescence lifetime and mitochondrial clustering. On the other hand, we use second harmonic generation (SHG) approach to image collagen fibers in a 3D context. We develop 3D orientation and organization algorithms to characterize the spatial alignment of collagen, which proves to be a sensitive biomarker in several diseases, such as cancer and traumatic brain injury. Overall, these improvements in cell and matrix characterization lead to a better understanding of cell-matrix interactions.


【報告人簡介】劉智毅,博士𓀚,2013年獲清華大學理學博士學位🦮,之後進入美國塔夫茨大學生物醫學工程系開展博士後研究至今🏊🏼‍♂️,導師為Irene Georgakoudi教授🧑。主要研究方向為生物醫學光子學⚇,包括雙光子熒光成像,熒光壽命成像,二次諧波成像等技術及在疾病診斷中的應用,並發展了相應的圖像處理方法和表征參量。已發表SCI論文20余篇,其中包括Biomaterials, Biosensors & Bioelectronics等第一作者或通訊作者的論文10篇;在合作作者論文中🎙,包括Science Translational Medicine 的封面文章等🏃🏻‍♀️‍➡️。擔任Chemical Communications, Journal of Biophotonics, Biomedical Optics Express等雜誌的審稿人。
